Transformation, Over Expression and Purification of Recombinant Antibody against Thyroid Cancer. • Carried out transformation of pET22(b) plasmid vector in Escherichia coli BL21 (DE3) cells for KT75 using electroporation. • Growth of the cells in Tartoffs-Hobbs media with appropriate antibiotics. • Induction of the KT75 protein using isopropyl β-D-1,thiogalactopyranoside(IPTG) • Harvesting of the induced cells and lysis of the cells to determine the protein location. • Dialysis of the overexpressed protein. • Purification from cell lysate using Ni-NTApurification. • Confirmation of the purified protein using SDS- polyacrylamide gel electrophoresis. • Protein labelling ie. Iodination of Thyroglobulin with I125. • Size Exclusion chromatography of labelled Thyroglobulin using Sephadex G-25 column. • Affinity determination of the antibodies towards Tg through Radioimmunoassay. Immunization of T7 Tail Fibre Antigen in Rabbit for Generation of Polyclonal Antibodies conducted in BARC. • Preparation of emulsion using Complete Freund’s Adjuvant. • Immunization of the rabbit using an emulsion prepared from purified protein. • Immunodiffusion performed for confirmation of antibodies against the T7 phages Show less
