Cell suspension cultures of chicory roots are used to product compounds of interest and study metabolic pathways. It has been demonstrated that the ratio between 5-mono-O-caffeoylquinicacid (5CQA) and 3,5-di-O-caffeoylquinic acid (3,5diCQA) observed in Chicory roots (Cichorium intybus var. sativum) extracts is reversed in cell suspension culture initiated from Chicory roots. In the latter, 3,5diCQA contents (6.24 µmol/g DW) are 3 fold higher than 5CQA (2.05 µmol/g DW). Thus this model could provide information about the biosynthetic pathway of 3,5diCQA which still need to be elucidated. For this purpose, we investigated conditions giving contrasting 3,5diCQA/CQA ratios by eliciting cells with methyljasmonate (MeJa), a known activator of the phenylpropanoid pathway. Phenolic acids were extracted and quantified using LC-DAD. At 50µM MeJa, the content of 3,5diCQA increased up to 26 fold and the content of 5CQA up to 40 fold, seven days after MeJa elicitation. Extracts were also analyzed by LC-ESI-MSn. Mass spectrometry data were analyzed to get more information about the evolution of global metabolism in presence of MeJa. We have completed our study with a comparison of the expression levels variations of 6 genes involved in metabolic pathway from phenylalanine to 5CQA (HQT, HCT, PAL1, C4H, C4L, C3H). The expression of 4 of these genes increased following the addition of Meja (HCT, PAL1, C4H, C4L). These results confirm this experimental system and its use for a deepest transcriptomic analysis.
