I had the opportunity to work with Antonia in my fall 2021 Science of Sales course, which is structured around a team of five students working together on a semester-long project to produce a professional-level sales plan for an existing company. Duplication mutations in the DMD gene account for 11% of Duchenne Muscular Dystrophy (DMD) cases. Currently, there is no cure available for these patients and care is restricted to symptom management. The Cohn lab has recently developed an innovative single guide RNA-CRISPR/Cas9 strategy to remove duplication mutations that was tested in vitro in cells derived from a DMD patient with a duplication from exon 18-30 in the DMD gene. To test our genome editing strategy in vivo, we generated the first multi-exonic DMD duplication mouse model (Dmd dup 18-30), which mimics a patient duplication from exon 18-30 in the Dmd gene. Molecular, histopathological and behavioural analysis confirmed that the Dmd dup 18-30 mouse recapitulates DMD patients phenotypes. We are now testing the efficacy of the single guide RNA strategy in removing the Dmd duplication in vivo, in the Dmd dup 18-30 mouse model. Show less
