In my Diploma Thesis I studied epigenetic marks of human tissue-specific gene promoters. I classified genes by differential expression patterns across 16 Human tissues samples (RNA-seq). On a genome-scale I characterized the so classified genes based on sequence characteristics and epigenetic modifications (ChIP-seq). To efficiently mine data, I developed tools in R and C++, such that I could test the hypothesis of epigenetic differences between housekeeping and tissue-specific gene regulation. Housekeeping genes are controlled by promoters which are ubiquitously initiated/accessible. Their gene transcription is regulated at the transition to transcriptional elongation. On the contrary, tissue-specific genes are controlled at the stage of transcriptional initiation. Show less
