The project focused on the verification of a novel EWSR1-BRCA1 interaction and its characterization in terms of transcriptional regulation and DNA repair through homologous recombination. Performed the cloning of plasmids, transformations, mini/maxipreps, PCRs, siRNA and plasmid transfections, co-immunoprecipitations, western blots, PCRs, cell culturing, immunofluorescent staining, fluorescent microscopy, and the generation of a CRISPR-Cas mediated EWSR1-/- cell line a.o. For the development of forensic anthropological identification methods, a contemporary and representative reference database of the Dutch population is required, one which is currently lacking. The set-up of a virtual database derived from patient CT scans could provide a solution, however, any methods derived from a virtual database should be valid on virtual bone models, as well as on physical dry bones, as the latter is most frequently used in forensic anthropology. For this reason, the accuracy of 3D virtual bone models of the os coxae, derived from patient setting CT-scans, when compared to its physical dry bone counterpart was tested. As a continuation of the bsc. internship project, immunolabeling and confocal imaging was performed of alpha- and beta microtubules and inositol phosphate 3 receptors (IP3) in atrial and ventricular myocytes. Additionally, to verify the expression of, and correct binding of antibodies to, IP3, western blots were performed. The aim of the study was to investigate possible alterations in the subcellular structural organization of X-ROS signalling in cardiomyocytes using an experimental animal model of tachy-paced induced atrial fibrillation. The most common techniques performed included the Isolation and fixation of atrial myocytes; membrane staining and confocal imaging of atrial cells with fluorescent dyes; and Immunolabeling and quantification of microtubule structures in intact cells.
