● Apart from learning and experimenting on different strategies to purify difficult proteins and crystallize them, I started planning and working on a relatively bigger project that involved molecular cloning of whole Rho family GTPases (about 20 proteins) of a pathogen and its activator.● With the help of my supervisor Dr. Sunando Datta, I wrote and secured a research grant from the Department of Biotechnology, Govt. Of India with a budget of ₹ 70 lacs.● I organized a team of 4 people which also included a JRF hired for this project. I learned key project management skills while working on this project as it included a large no. of reagents purchases, meticulous planning, time management, cost management, and best possible communication and interpersonal skills. My team managed to procure reagents, design primers, amplify genes using PCR, develop more than 40 plasmid constructs, express about 21 proteins, purify and validate functionality using HPLC and perform GEF screening assay using spectrofluorimetry. All this was achieved in about 5 months.● I also learned and gained expertise in cell culture when I expanded my in vitro observations to Entamoeba histolytica (a human pathogen). My cell culture experiments allowed me to gain expertise in transfection, western blotting, immunofluorescence assay, confocal microscopy and development of different functional assays.● Some of my experiments involved protein interaction studies which allowed me to gain experience in Isothermal Titration Calorimentry (ITC).● Discoveries and data from this project motivated me to write a second research grant. Foreseeing the requirement of an expert on encystation (an amoebic life cycle process), we collaborated with Dr. Swati Tiwari from Jawarharlal Nehru Univ., New Delhi. I wrote the second grant in collaboration and secured ₹1 crore under Scheme for Transformational and Advanced Research in Sciences (STARS).

● During my work as a JRF, I learned molecular biology techniques such as PCR, molecular cloning, protein expression, protein purification using affinity chromatography, size exclusion chromatography, ion-exchange exchange chromatography.● I also learned various protein crystallization methods such as the hanging drop vapor diffusion method and batch method. I also learned to utilized various protein crystal optimization strategies such as matrix screening, dilution method using vapor phase diagram.● During my early days as a JRF, I learned how to do a literature search, data mining, presentation of technical data. My skills as an effective communicator and presenter were sharpened throughout my work as we had a weekly one to one presentation as well as bimonthly group presentation.
