  RESEARCH PURPOSE California’s state lichen, Ramalina menziesii, is not only a well recognized symbiont, it has also been discovered to contain high concentrations of monomethyl mercury (MMHg) along Northern California’s coastal regions. It is considered an important atmospheric biomarker based on its ability to bioaccumulate atmospheric. The abundance of the MMHg in these coastal species raises the question of Ramalina menziesii’s role in it’s deposits. Does this lichen species reflect high MMHg atmospheric pollution through bioaccumulation or is it contributing to MMHg production? The following article discusses the motivation, methods, and results of the sequencing of Ramalina menziesii’s metagenome to discover the lichen’s ability to methylate mercury. METHODS 1) Purified high molecular weight DNA of Ramalina menziesii using the Promega Wizard Genomic DNA purification kit 2) Phenol-chloroform extractions to further remove impurities and proteins 3) Illumina Tn5 transposase library build with HISeq 1200 Illumina Sequencing 4) MinION Nanopore Sequencing 5) Genome Assembly with Shasta, polishing with Pilon 6) Metagenome analysis with MG-RAST 7) HgcAB gene search accomplished with bioinformatics pipeline (HMMER, Glimmer, Clustal Omega) Show less
