      Applied site-directed and random mutagenesis techniques to identify polymorphisms to develop desired traits. Utilize various expression systems such as E. coli, S. cerevisiae, A. tumefaciens, and A. thaliana. Screened libraries utilizing various molecular methods such as transient expression in N. benthamiana, colorimetric-based protein activity assays, in-gel activity assays and Yeast-Two-Hybrid- Characterized and validated induction of gene(s) in plant species against bacterial/fungal infection-- Employed site-directed mutagenesis (SDM) based on computational recommendations to change the enzymatic activity of genes of interest.-- Screened 30+ SDM mutants using transient expression in N. benthamiana for qualitative and quantitative assays that detects activity.-- From the mutations generated, identified polymorphism in gene of interest that changes the protein structure and functionally changes the enzymatic activity, which led to a capture of company IP.-- Identified mutations have been recognized as a novel gene editing trait.- Created an in-house multi-site saturated library of mutations (over 8,000 variants) and QC’d library with NGS.- Conducted random mutagenesis to generate a library of single nucleotide polymorphisms into gene of interest and evaluate for strong protein-protein interaction in Yeast Two-Hybrid screening.-- Identified 20 candidates from Yeast Two-Hybrid screening -- Performed transient expression in N. benthamiana for in planta validation of gene against pathogen effectors and found one candidate with desired effects when challenged with the effector.-- Confirmed expression of transiently expressed proteins in N. benthamiana using SDS-PAGE and Western Blot.- Supervised and mentored junior staff and wrote monthly and quarterly progress reports, present results and data to small research group as well as the company’s scientific team.
