      Topic: The impact of early life antibiotics on gut maturation Antibiotics are the most commonly prescribed medication in pediatric care, both to prevent and treat infectious diseases. There are, however, several diseases associated with the prolonged use of early life antibiotics. I aimed to investigate the effects of early life antibiotic treatment on intestinal development after birth. Main projects: • establish, optimize and characterize an in vitro model to study intestinal postnatal maturation using mouse fetal intestinal organoids (published in EMBO Reports). • video protocol of mouse fetal intestinal organoids model (published in JOVE). • study the direct effects of antibiotics on the intestinal cells, by comparing the treatment in young mice with the treatment of mouse fetal intestinal organoids (under revision). • determine the long-term consequences of early life antibiotic treatment on the adult intestine (in preparation). • optimize and characterize the culture of human fetal intestinal organoids (to be finished by next PhD candidate). Responsibilities: • work in ISO 15189 accreditation environment. • conduct experiments using young mice, fetal and adult/mouse and human/2D and 3D intestinal organoids, qRT-PCR, immunostainings, seahorse metabolic assays, enzyme activity assays, microarray analysis, FACS and ELISA. • obtain approval for a 5-year project from the Dutch Committee for Animal Research (Aanvraag Projectvergunning Dierproeven), including concept creation, experimental planning, protocol development, and resource planning. • closely collaborate with researchers within my group and the department, and from other groups in the hospital and external partners. • present results in several national and international conferences, for both scientific and lay audiences. • train other researchers in certain techniques. • supervise 3 students in thesis and project proposal writing and presenting. • organize biweekly literature discussions.
