My previous research project involved working on understanding the unique relationship between human neurons and varicella zoster virus in a pure neuronal population in vitro. The goal was to establish a model of latent in vitro infection in order to study reactivation of the virus for shingles prevention research. Using immunocytochemistry, quantitative PCR, paired-end directional RNA sequence analysis, and western blot we investigate viral and cellular pathways influenced during lytic and non-lytic infections in vitro. Conducted preliminary research on orally administered malaria vaccines with Dr. Susan M. Keenan. Briefly, we identified three antigenic targets of Plasmodium (P.) falciparum, one antigenic target per lifecycle stage. Specifically, Sporozoite Surface Protein 2 (SSP2) from the liver stage, Merozoite Surface Protein 1-19 (MSP1-19) from the blood stage, and Plasmodium falciparum surface protein 48/45 (Pfs48/45) from the sexual stage of infection. Using standard cloning techniques we generated four plant expression vectors containing these antigenic targets of P. falciparum individually and combined. To show proof of concept and vector functionality, we genetically engineered Agrobacterium tumefaciens to contain the expression vector containing the liver stage antigenic sequence. Finally, utilizing the modified A. tumefaciens we transformed the model plant Arabidopsis thaliana and showed the expression of the liver stage antigen SSP2 through western blot analysis of the plant matter. To summarize, we demonstrated that A. thaliana derived SSP2 protein was recognized by human antibodies to P. falciparum SSP2 antigen. The project gave proof of concept that plants are capable of producing P. falciparum proteins that are recognized by human derived antibodies to P. falciparum, providing preliminary data for future malaria vaccine research. I investigated the role of the rice FtsH3 protease in biomass accumulation to guide future improvements of more intractable bioenergy crops under Dr. Jan Leach at Colorado State University. We silenced the FtsH3 protease in rice seeds using genetically engineered Agrobacterium tumefaciens and studied the phenotypic results as well as identify single nucleotide polymorphisms in FtsH3 using genomic sequencing techniques.
