In our research project that is funded by Merck I am developing a novel in-vitro model using organoid and organ-on-a-chip technology. We want to understand the connection of the intestine in the development of autoimmune diseases. Expertise highlights: - processing of whole organ transplants - cell isolation from primary, fresh or frozen tissue - organoid generation from all different intestinal regions - organoid manipulation to fit assay requirements - assay design and establishment to study intestinal epithelial barrier - physiological and disease specific pathophysiological modeling - analysis and interpretation of complex RNA sequencing data from divers patient population - preparation of single-cell RNA sequencing libraries Chairing the Special Interest Group of Career Development where we are organizing presentations and workshops. In my PhD project I have established an intestinal 3D culture (organoids) in our lab. I have introduced, modified and improved known protocols to grow organoids in defined differentiation stages. The success of the differentiation was confirmed using marker gene expression analysis, histologic stainings and immunhistochemistry. Furthermore the cultures were used to study ion transporters in the different intestinal cell types e.g. by a swelling assay. Furthermore monolayer cultures of the organoids on different permeable supports were established to measure ion transport using Ussing chamber techniques and microfluorometry. Show less
