      Characterizing of a novel class of chromatin modifiers which interfere with cancer metastasis.In this project we are using: CRISPR-Cas9 system to generate KO's, ChIP-seq, RNA Micro-arrays and SILAC LC-MS proteomics approaches.

      During my PhD project I studied the role of post-translational modifications of certain fibrosis and cancer related processes. Of particular interest was understanding the molecular functions and transcriptional (epigenetic) regulation of genes related to collagen deposition and cross-linking. Amongsts other things, I discovered novel interactions of enzymes that are important for the inititation of collagen cross-linking (PNAS). Also, I dissected the trancriptional regulation of collagen cross-linking gene PLOD2 in fibrotic fibroblasts (JBC). Furthermore, I used "state of the art" gene targeting tools (Zinc Fingers and CRISPR-dCas9) to initiate epigenetic editing as a manner to interfere with target gene expression (Nature Communications).This research was funded by the NIRM (Netherlands Institute for Regenerative Medicine), and in practice a collaboration of the labs of Prof. Dr. Ruud Bank (Tissue Generation & Repair group) and Prof. Dr. Marianne Rots (Epigenetic Editing group).My PhD was awarded with highest Dutch academic distinction.Click the link below to read my PhD thesis online.
