I am responsible for maintaining and running a high throughput departmental NGS facility. I am familiar with a wide range of NGS equipment and techniques for everything from the size selection of libraries, to library preparation, to sequencing. My most commonly fulfilled sequencing requests include ChIP-Seq and RNA-Seq experiments. My contributions to NGS analysis does not stop at the bench. I am also familiar with managing the large amounts of data we generate and have a working knowledge of what it takes to transition files from .bcl to .fastq, .bam, .bw and onwards to EdgeR, MACS, and SICER. My project aims to combat melanoma by targeting the telomerase and telomeres of cancer cells. Telomeres are the caps of repetitive TTAGGG sequences on the ends of chromosomes that act as a safety region and prevent the chromosome from shortening every time our cells divide. Telomerase is a protein that aids in protecting these telomeres. Therefore, we have hypothesized that the drug T-oligo and 6-thio-DG will prevent telomerase from protecting telomeres in cancer cells and eventually cause apoptosis or cell death by induced DNA damage through chromosome shortening Some of the requirements expected of my position: •Assisting the faculty writing NIH grant and helping the funding of the lab •Assisting colleagues with their academic research •Maintaining various melanoma cancer cell lines •Assisting the faculty with scientific writing •Attend project meetings •Analyzing data. I also work on a novel photodynamic therapy with a novel drug called MP106 and a well-known 5-ALA drug in combination with a 405nm LED to treat the MMAN melanoma cancer cell line. The techniques used here were: •MTT cytotoxicity bioassay, study the drug binding mechanism, & western blot analysis •Culture and maintenance of aseptic mammalian cells in multiple formats, cryopreservation, cell counting, & cell lysate preparation •Additional laboratory rotation helped in learning in vitro cellular cell migration and transformation assay, & immunohistochemistry. Currently as of May 2018, I have began research on NSCLC and osimertinib. Osimertinib is a third-generation, oral, irreversible epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor. Osimertinib exhibits a nine-fold greater affinity for select EGFR-sensitizing and T790M-resistance mutations than to wild-type EGFR. In vitro, osimertinib inhibits the activity of HER2, HER3, HER4, ACK1, and BLK. Show less
