• Cell culture including: isolation of primary bovine retinal microvascular endothelial cells and primary murine brain-derived microglia, cell lines (HEK293A, THP-1 and BV-2 monocyte/macrophage), cell culture in hypoxia and hyperoxia. • Oxygen Induced Retinopathy mouse model • Aortic ring assays • Adenovirus preparation and purification, and viral infection of primary cells. • Cell/ tissue preparation for immunohistochemistry • Retinal flat mounts • Viability(LDH assay)/apoptosis (nuclear assessment, active caspase-3 staining, ApoToxGLo assay)/senescence (γH2AX, 53BP-1, SAβ-G) detection optimisation and quantification • Detection and measurement of reactive oxygen species (DHE assay) and reactive nitrogen species (3-Nitrotyrosine western blotting and Griess assay) • Light, fluorescent and confocal microscopy • Protein extraction for SDS PAGE/western blotting • RNA extraction, cDNA synthesis, PCR and gel electrophoresis • Image and data analysis using software including:EZ-C1, Fiji, NIS Elements, Excel, Graph-pad Prism • National and international conference publications and presentations, chairing seminars • Mentor undergraduate students with lab work • Acquiring travel funding for national and international conferences, lobbying for funding for lab equipment, expenditure accounts
