I prepared dissociated organoids in transwells for SARS-CoV-2 inoculation and and drug screening assays. I oversaw virus production and expansion of human Coronavirus OC43 and 229E and infection of lung organoids in air-liquid interface culture. I also isolated human gastrointestinal organoid lines from Crohn’s Disease and Ulcerative Colitis patient biopsies, cultured dissociated organoids in transwells and took transepithelial resistance measurements I created new myofibroblast cell lines, designed a multicellular model of fibrotic Crohn’s disease, and performed assays to optimize growth media. As a cell culture technician, I am responsible for isolating, culturing and harvesting, and performing quality control assessments of primary cell lines. I have worked extensively with human GI organoids and neural stem cells, as well as over 20 different human, rat and monkey neuronal cell types. I have optimized microdissection and isolation techniques for 7 different types of embryonic rat neurons. I have also gained experience in RNA isolation and characterization via gel electrophoresis and immunocytochemistry and fluorescent imaging.
